It would help if I knew what panel you use. I had the same problem with a 727 in FS2004 and the problem was in the aircraft cfg. file. But without knowing which panel you are using I am shooting in the dark ???.
Try pasting a screenshot of it and I might be able to help you. The 727 is one of my favorite planes.

autopilot on the 727?

Okay skipper, compared to modern jets, the 727 is aged when it comes to autoflight systems.
You'll find no autothrottle (hence no IAS or mach hold), can't pre-select altitude and vertical speed, just a  simple altitude hold. It has a heading select and VOR course select function, as well as ILS capture ability.
Umm...yeah, that's about it. Old school flying that requires lots of attention and skill....I suggest you read the MANUALS if there is one available.

OK MUZ I looked at the panel you referred to and I can help you. I use to use this panel before I got the one I now have. I have to agree this panel is not like any other panel you might see, but the 727 is not really like any other plane either and this panel closely resambles the real thing.
If you look on the right side of the panel you will notice two black levers side by side the left one arms the auto pilot and the right is the auto throttle to set the speed you need to click on the speed bug on the airspeed gauge lower right hand corner if you put your coursor on one side you will see a plus and a minus on the other. The black knob right in the upper middle is where you set your modes at HEADING VOR LOCK(nav hold) APPROACH or BACKCOURSE the tricky part of this panel is how you adjust how you control your rate of climb or descent close to the airspeed indicator you will see a knob with a airplane on it this is what that is used for.
Do not try to take off using auto pilot as it will not work
Its better to take off manually then set the auto pilot.
One trick I learned is that if you go to any other plane and set the auto pilot there then switch back to the 727 all of the settings will transfer over. Inotherwords it will be set for you. By doing this you can see what switches or set.
